Description of project idea:
We are a consortium made of 4 partners aiming at developing nanoparticols for delivering anti-diabetes drug targeting epigenetic mechanisms specifically in the adipose tissues. The coordiantor of. the consortium has demonstrated that inactivation of Suv420h histone methyltransferases specifically in mouse adipose tissue improve metabolic parameters and is protective toward diet-induced obesity. We want to take advantage of nanotechnology for generating nanoparticles containing a validated Suv420h inhibitor for diabetes treatment.

Description of project idea:
Autoimmune thyroid diseases (ATD) and Type 1 diabetes (T1D) are the most common autoimmune endocrinopathies. The combination is polyglandular syndrome Type 3 variant (APS3v). The PTPN22 C1858T variant allele, encoding for R620W Lyp, is frequently found associated with these diseases. Lyp is a negative regulator of TCR signaling. Lyp variant leads to a gain of function activity with paradoxical reduced T cell activation. We achieved in vitro downregulation of PTPN22 and its variant using liposomes complexed with short interfering RNA duplex (siRNA) (lipoplexes). Systemic inhibition of PTPN22 variant can be developed as a novel drug for prevention or early immunotherapy for T1D and APS3v patients with the variant. We aim to: 1) prove the efficacy/tolerability of functionalized lipoplexes in prevention/treatment by injection in mice NOD harboring the PTPN22 variant 2) tailor and test the best-targeted liposomal carrier to improve systemic delivery/kinetics 3) test the effect of lipoplexes on human inflammed islets harbouring the PTPN22 variant in vitro 4) produce clinical grade complexes.

Description of project idea:
We are solving a well-known problem in the ophthalmic industry. Less than 5% of the applied eye drop reaches the eye. Most topically administered drugs are washed away within a few seconds after instillation because of blinking and tears discharge. In terms of the product, we have succeeded to incorporate specific nanoparticles into regular hydrogel material, and voila - there is a new material with super-absorptive properties with awesome ability to normalize the pH level of the eye. The lenses absorb bacteria, toxins, chemicals, and possess anti-inflammatory and antioxidant properties. Recently have successfully passed a safety test with the clinical research organization LEITAT in Barcelona, and the efficacy test for antioxidant properties gaining excellent results.

Doxorubicin is the most common chemotherapeutic agent used for numerous cancer treatments by induction of cancer cell senescence. The senescence of cancer cells is an excellent approach to stop cancer cell growth, but that senescent cells, if it remains without elimination, will proliferate and causing cancer relapse. Therefore, senolysis of senescent cancer cells is a new and novel approach to increase the efficiency of doxorubicin and its safety by the protection of the normal cells from the side effects of doxorubicin. Herein, we will investigate the senotherapeutic effect of some natural bioactive compounds (BAC)-NPs including (thymoquinone, curcumin, caffeine, diosgenin, rutin, quercetin, genistein, and fisetin) on the senescent liver, breast, and colon cancer cells.

As soon as we form a consortium for the EuroNanoMed3 proposal, As a coordinator I would like to develop CRISPR-Cas and aptamer based miniaturized electronic diagnostic devices. These miniaturized devices will be capable of detecting liquid biopsy markers such as circulating tumor cells (CTCs) and circulating tumor DNAs (ctDNA) and RNAs. In this proposal, CRISPR-Cas system will detect tumor DNAs and RNAs. Aptamer based system will also detect CTCs. The whole system will detect tumor heterogeneity, mutation, origin of the tumor and metastasis. All in all the diagnostic system is able to monitor diagnosis, prognosis and the treatment progress by measuring these materials. That is I can provide for the technology development. Our company develops synthetic polypeptide-based materials starting from natural amino acids. We use N-carboxyanhydride polymerization (NCA) technique to obtain poly(amino acid)s i.e. synthetic polypeptides. NCA polymerization is also the method for the producton of the famous multiple sclerosis (MS disease) complex drug called copolymer-1 (i.e. glatiramer acetate, Copaxone). Recently, we combined NCA technique and recently explored triazolinedione click chemistry to obtain hydrogels and films from synthetic polypeptides. NCA technique is advantageous over solid phase peptide synthesis (SSPS) in terms of scalability, cost and polymer lenght. On the other hand, TAD chemistry is advantageous over other click chemistry reactions as this reaction does not require catalyst, UV, heat or byproducts.
We specifically focus drug delivery, RNA delivery and tissue engineering applications as our materials will have superior advantage over traditional polymers and hydrogels. We can provide products with varying range of amino acid composition, different mechanical properties and tunable porosity of the hydrogel network.

The title: A nucleotide-modified RNA-aptamer, selected to the self-surface of human B-cell lymphoma cell, as the targeting moiety for delivery of therapeutic molecules.
We have generated nucleotide-modified RNA-aptamers (nmRNA-aptamers) (80 - 100 nt oligonucleotides) that recognize the cell-surface molecules of malignant B-cell lymphoma (Burkitt lymphoma, BL) cells, using the Whole-Cell SELEX method modification (Cell-SELEX-FA) elaborated by us previously. Viability tests have demonstrated that one of the selected nmRNA-aptamers (at concentration 4.0 µM) inhibited cell proliferation of the all studied highly malignant CD10+ BL lines (n=5) from 56% up to 99%. The same nmRNA-aptamer (nmRNA-apt) was inactive in T-cell lymphoma cell lines and normal donor lymphocytes. Soft-Agar-Growing Test has also demonstrated significant inhibition of the cell-colony formation of the CD10+ BL lines by this nmRNA-apt. We assumed that the selected nmRNA-aptamer can be suggested as the carrier molecule to target malignant B cells for delivery of therapeutic molecules.
The aim of the WP#: investigation of potential therapeutic application of the generated nmRNA-aptamer as the targeting moiety for delivery of therapeutic molecules in human B-cell lymphoma model.
The methodology will include the study: (1) using the panel of human high-malignant B-cell lymphoma cell lines (BCLs) versus the EBV-immortalized non-malignant lymphoblastoid B-cell lines (LCLs); (2) on the panel of human cancer cell lines of different tissue origin and (3) using the peripheral blood samples from healthy individuals and patients with B-cell lymphomas, diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma (FL).
Expected results of the WP#: to work out the protocol (dozes per malignant cell, input incubation time-schedule) by which both the highest BCL cell-proliferation blocking effect and the lowest toxic effect for non-malignant B cells will be achieved in order to suggest the new developed strategy, based on the application of the nmRNA-aptamer complex and the elaborated protocol, for the treatment of patients with high-grade aggressive B-cell lymphomas.
